Insuman Comb 30

Insuman Comb 30 is a biphasic human insulin suspension for injection, consisting of 30% regular human insulin (short-acting) and 70% NPH (Neutral Protamine Hagedorn) insulin (intermediate-acting). Developed by Sanofi, this formulation is designed to mimic the physiological insulin response by providing a rapid onset of action to cover mealtime glucose excursions and a prolonged duration of action to maintain basal glucose levels. The regular insulin component acts by binding to the insulin receptor, stimulating peripheral glucose uptake in skeletal muscle and fat, while simultaneously inhibiting hepatic glucose production. The NPH component, which is insulin complexed with protamine, slows absorption to extend the therapeutic effect. In the LAPTOP (Lantus Plus Amaryl Plus Metformin Versus Premixed Insulin) clinical trial, Insuman Comb 30 was utilized as a twice-daily comparator to evaluate the efficacy of insulin glargine-based regimens in patients with type 2 diabetes mellitus who were inadequately controlled on oral antidiabetic agents.
