Overview

INT-1B3 is a lipid nanoparticle (LNP)-formulated, chemically modified synthetic mimic of the endogenous tumor suppressor microRNA miR-193a-3p. Developed as a novel microRNA-based therapeutic, it is designed to address multiple hallmarks of cancer simultaneously by modulating both tumor cells and the tumor microenvironment. Mechanistically, INT-1B3 acts by restoring miR-193a-3p function, leading to inhibition of proliferation and migration, induction of cell cycle arrest and apoptosis in cancer cells, and triggering immunogenic cell death. It also downregulates the adenosine-A2A receptor pathway via inhibition of CD39/CD73, resulting in decreased immunosuppressive regulatory T cells (FoxP3/Lag3) and monocytic myeloid-derived suppressor cells (mMDSCs), while promoting dendritic cell maturation and recruitment of CD8+ effector T cells. This results in enhanced anti-tumor immunity with reduced metastasis development[4][5][6][7][8].
