Overview

INTASYL is a proprietary self-delivering RNA interference (RNAi) platform developed by Phio Pharmaceuticals. It enables the creation of uniquely modified small interfering RNA (siRNA) compounds that can efficiently enter target cells without the need for specialized drug delivery systems or formulations. The technology is designed to silence specific genes—such as those encoding immune checkpoint proteins like PD-1 and CTLA4—that tumors use to evade immune detection and destruction. By silencing these targets in immune cells (e.g., T cells and natural killer cells), INTASYL compounds enhance the anti-tumor activity of these cells and improve outcomes in immuno-oncology therapies. Lead product candidates include PH-762 (targeting PD-1), PH-894 (targeting BRD4), and others targeting Cbl-b or MMP1 for various indications including cancer immunotherapy and skin disorders such as photo-aging[2][3][6][9].
