An integrin αvβ3 blocking antibody is a monoclonal antibody designed to target the αvβ3 integrin, a cell surface receptor that plays a critical role in cell-extracellular matrix (ECM) interactions, angiogenesis, and tumor progression. Integrin αvβ3 is typically overexpressed on the surface of activated endothelial cells in the tumor neovasculature and on various malignant cells, including those in glioblastoma and melanoma. By binding to the αvβ3 heterodimer, these antibodies block its interaction with RGD-containing ligands such as vitronectin, fibronectin, and L1CAM. This blockade disrupts downstream signaling pathways (e.g., FAK, Src, and PI3K/Akt) that are essential for cell adhesion, migration, and survival. Historically, several therapeutic candidates have been developed in this class, including etaracizumab (Abegrin) and abituzumab, primarily for the treatment of solid tumors and bone metastases, although many have faced challenges in clinical trials due to limited monotherapy efficacy.
