Integrin beta-3-neutralizing antibody is an experimental monoclonal antibody designed to target and inhibit ITGB3 (Integrin beta-3), a cell surface receptor identified as a critical regulator of T cell exhaustion in solid tumors. In esophageal squamous cell carcinoma (ESCC), ITGB3 expression on T cells marks a transitional state toward terminal exhaustion, characterized by reduced cytotoxicity and impaired proliferation. By neutralizing ITGB3, the antibody restores T cell effector functions, including the secretion of interferon-gamma (IFN-γ) and granzyme A, and enhances the tumor-killing capacity of the immune system. Preclinical studies demonstrate that this antibody can reduce tumor growth as a monotherapy and exhibits synergistic effects when combined with PD-1 inhibitors, effectively overcoming resistance to immune checkpoint blockade by diminishing the exhausted T cell pool.
