An integrin-binding antibody is a type of monoclonal antibody designed to target and bind to integrins, a family of transmembrane heterodimeric cell surface receptors composed of alpha and beta subunits. Integrins play critical roles in cell-extracellular matrix (ECM) adhesion, cell-cell interactions, and intracellular signaling pathways that regulate cell survival, proliferation, migration, and angiogenesis. In oncology, integrin-binding antibodies (such as those targeting integrin alpha-V/beta-3 or alpha-V/beta-5) are investigated for their ability to inhibit tumor angiogenesis, disrupt tumor cell adhesion and migration, and prevent metastasis. Additionally, research has demonstrated that targeting the integrin alpha-V/beta-3/HSPG complex can block the endocytosis-mediated uptake of cancer-derived extracellular vesicles (EVs), thereby halting intercellular communication that promotes pre-metastatic niche formation.
