Interferon-β + retinyl palmitate + tamoxifen is a combination therapy that leverages the complementary mechanisms of three distinct agents to treat metastatic breast cancer. This regimen combines: 1. **Interferon-β**: A type I interferon with immunomodulatory and antiproliferative properties that upregulates MHC I proteins and enhances CD8+ T cell activation. 2. **Retinyl palmitate**: A retinoid (vitamin A derivative) that potentiates interferon effects by increasing levels of STAT1, STAT2, and interferon regulatory factors. 3. **Tamoxifen**: A selective estrogen receptor modulator that blocks estrogen activity in breast tissue by competitively inhibiting estrogen binding to its receptor. The combination has demonstrated additive or synergistic antiproliferative effects on breast cancer cell lines. Clinical studies have shown that long-term administration is feasible with moderate toxicity, and the regimen has shown activity in pretreated metastatic breast cancer patients regardless of estrogen receptor status.
