Overview

This is a combination therapy consisting of interferon alfa (a type I interferon cytokine) and tamoxifen (a selective estrogen receptor modulator). The rationale for combining these drugs is based on preclinical and clinical evidence suggesting that interferon alfa can increase the expression of estrogen receptors in breast cancer cells, potentially enhancing the sensitivity of these cells to the growth-inhibitory effects of tamoxifen. Tamoxifen acts primarily by competitively inhibiting estrogen binding to its receptor, thereby blocking estrogen-driven tumor growth. Interferon alfa exerts antiproliferative and immunomodulatory effects. Clinical studies have explored this combination mainly in advanced or metastatic breast cancer, with some reports indicating additive antitumor activity but also increased risk of hematologic toxicity such as leukopenia and thrombopenia[1][2].
