Overview

Interferon alpha-4 (IFNα4) is a specific subtype of type I interferons (IFN-Is), a family of cytokines crucial for modulating antitumor immunity. Preclinical studies in murine models have shown that IFNα4 can temporarily restrict tumor outgrowth. However, it also drives an intense, yet often ineffective, IFN-I response that ablates type 2 inflammatory cells within the tumor microenvironment, ultimately leading to defective T cell immunity. Research indicates that this specific imbalance, characterized by high IFN-I activity and low type 2 inflammation, is associated with poor clinical outcomes across various solid cancers.
