Overview

interferon-gamma gene therapy (DNAtrix) is an investigational oncolytic immunotherapy designed for the treatment of glioma. Developed by DNAtrix, the therapy (often referred to by the code name DNX-2411) utilizes a modified oncolytic adenovirus (Delta-24) as a vector to deliver the human interferon-gamma (IFN-γ) gene directly into the tumor. The virus is engineered to replicate selectively in Rb-pathway deficient cancer cells, leading to direct cell lysis. Simultaneously, the expression of the IFN-γ transgene within the tumor microenvironment is intended to stimulate a robust anti-tumor immune response by enhancing antigen presentation, activating natural killer (NK) cells and T cells, and reversing local immunosuppression. This approach aims to provide a localized, high-concentration delivery of IFN-γ to the brain while minimizing systemic toxicity.
