Interferon gamma-primed human bone marrow-derived mesenchymal stromal cells (γMSCs) are an investigational allogeneic cell therapy being developed for the prophylaxis of acute graft-versus-host disease (GVHD) in patients undergoing hematopoietic cell transplantation (HCT). The product consists of mesenchymal stromal cells (MSCs) harvested from human bone marrow that are expanded and licensed or primed with interferon gamma (IFNγ) prior to administration. This priming activates the IFNγ–JAK–STAT1 signaling axis, inducing the expression of potent immunosuppressive mediators including indoleamine 2,3-dioxygenase (IDO-1), interleukin-10 (IL-10), programmed death-ligand 1 (PD-L1), and prostaglandin E2 (PGE2). These factors collectively suppress T-cell proliferation and inflammatory cytokine production, aiming to prevent the onset of GVHD in patients treated for hematologic malignancies such as acute leukemia and myelodysplastic syndrome (MDS).
