Recombinant human interferon lambda-1 (IFN-λ1), also known as interleukin-29 (IL-29), is a member of the Type III interferon family developed by Nanogen. It functions as an agonist of the Type III interferon receptor, a heterodimeric complex composed of the IFNLR1 (IL-28Rα) and IL10RB subunits. Upon binding, it activates the JAK-STAT signaling pathway, leading to the phosphorylation of STAT1 and STAT2 and the induction of interferon-stimulated genes (ISGs) that provide antiviral, antiproliferative, and immunomodulatory activities. Because the Type III interferon receptor is primarily expressed on epithelial cells and is absent on most hematopoietic cells, interferon lambda-1 is hypothesized to have a more localized effect and a superior safety profile compared to Type I interferons (like interferon-alpha). It is being investigated for the treatment of various viral infections, including hepatitis B, hepatitis C, and respiratory viral diseases.
