Interleukin-10 (IL-10) mRNA nanoparticles are a preclinical therapeutic platform that utilizes lipid nanoparticles (LNPs) to deliver messenger RNA encoding the anti-inflammatory cytokine IL-10. This approach is designed to overcome the pharmacokinetic limitations of recombinant IL-10 protein therapy, such as its short systemic half-life and poor accumulation in target tissues, by enabling the patient's own cells to produce the cytokine in situ. In preclinical models of intracerebral hemorrhage, these nanoparticles—often targeted to the inflamed brain endothelium via VCAM-1—have demonstrated the ability to reduce neuroinflammation and improve functional outcomes. In oncology, specifically hepatocellular carcinoma, systemic delivery of IL-10 mRNA nanoparticles has shown potential in modulating the tumor microenvironment to induce immune surveillance and inhibit tumor growth.
