Interleukin 12 engineered dendritic cells are a form of autologous or allogeneic dendritic cell therapy genetically modified or otherwise engineered to express or secrete interleukin-12 (IL-12), a potent immunostimulatory cytokine. The engineered dendritic cells are designed to enhance antitumor immunity by activating both innate (e.g., natural killer cells) and adaptive immune responses (e.g., cytotoxic T lymphocytes) via the secretion of IL-12, leading to increased production of interferon gamma (IFN-γ), promotion of Th1 immune polarization, and attenuation of immunosuppressive regulatory T cell activity. The approach is under investigation for use in cancer immunotherapy, particularly for solid tumors such as glioblastoma and in vaccine strategies for other malignancies, with evidence of increased antigen presentation, T cell activation, and antitumor efficacy in preclinical and early clinical settings[1].
