Overview

Interleukin-12 (IL-12) gene therapy is an immunotherapeutic approach designed to induce the local expression of IL-12, a potent proinflammatory cytokine, within the tumor microenvironment. IL-12 plays a pivotal role in bridging innate and adaptive immunity by activating natural killer (NK) cells and cytotoxic T lymphocytes, promoting Type 1 T helper (Th1) cell differentiation, and stimulating the production of interferon-gamma (IFN-γ). The delivery of the IL-12 gene, typically via plasmid DNA or viral vectors, aims to stimulate a robust anti-tumor immune response while mitigating the systemic toxicity associated with the administration of recombinant IL-12 protein. A specific plasmid DNA construct, identified as NSC 709933, was developed and evaluated by the University of Wisconsin-Madison in Phase I clinical trials for the treatment of spontaneous skin metastases from various solid malignancies.
