Interleukin-12 plasmid deoxyribonucleic acid lipid nanoparticle (pDNA-IL-12 LNP) is an experimental gene therapy formulation designed to deliver a DNA plasmid encoding the interleukin-12 (IL-12) cytokine. IL-12 is a potent immunomodulatory cytokine that bridges innate and adaptive immunity by activating natural killer (NK) cells and inducing the differentiation of CD4+ T cells into Th1 cells, which are critical for anti-tumor responses. The lipid nanoparticle (LNP) component serves as a delivery vehicle, protecting the DNA from enzymatic degradation and facilitating its uptake into target cells, such as those in the liver or the tumor microenvironment. Once inside the cell, the plasmid is transcribed and translated to produce IL-12 protein locally. While effective in preclinical models of hepatocellular carcinoma (HCC) for suppressing tumor growth and reprogramming the 'cold' tumor microenvironment, pDNA-LNP formulations are often associated with significant systemic toxicity and off-target hepatic inflammation, which has led to the development of alternative platforms like circular single-stranded DNA (cssDNA).
