Overview

Interleukin-12 (IL-12) plasmid DNA is a genetic adjuvant developed by the National Institute of Allergy and Infectious Diseases (NIAID) for use in therapeutic and preventive HIV vaccine regimens. It consists of a circular DNA plasmid containing the genes for human IL-12 (p35 and p40 subunits). When administered, typically via intramuscular injection followed by in vivo electroporation, the plasmid enters host cells which then express and secrete the IL-12 cytokine. IL-12 acts as a potent immunomodulator that enhances the magnitude and quality of cellular immune responses, particularly Th1-type T-cell responses, against co-administered HIV antigens (such as those in the HIV-MAG DNA vaccine).
