Interleukin-15 DNA refers to a gene therapy or DNA-based therapeutic approach in which a plasmid encoding the human interleukin-15 (IL-15) cytokine is delivered to cells. The goal is to induce endogenous production of IL-15 protein, a cytokine with structural similarity to interleukin-2 that plays a critical role in the development, survival, and activation of natural killer (NK) cells and CD8+ T cells[3][4][5]. Mechanistically, expressed IL-15 binds its high-affinity receptor complex (IL-15Rα/IL2Rβ/γc), activating JAK1/JAK3 kinases and downstream STAT3/STAT5 signaling pathways[6][8]. This results in enhanced proliferation and anti-apoptotic effects on immune effector cells. In cancer immunotherapy research, delivery of IL-15 via plasmid DNA aims to boost anti-tumor immunity by increasing NK cell cytotoxicity and T cell responses[5]. The approach is under investigation for various cancers but has not reached regulatory approval.
