Interleukin-15 receptor alpha-Fc (IL-15Rα-Fc) is a recombinant fusion protein consisting of the extracellular domain of the IL-15 receptor alpha subunit linked to an immunoglobulin Fc region. This molecule serves as a therapeutic platform designed to enhance the activity and stability of the cytokine interleukin-15 (IL-15). In its natural physiological role, IL-15 is trans-presented by IL-15Rα on the surface of dendritic cells to the IL-2/IL-15 receptor beta-gamma complex on NK cells and CD8+ T cells. The IL-15Rα-Fc fusion protein acts as a soluble mimic of this process. When complexed with IL-15 or its variants, the Fc domain provides extended circulatory half-life through FcRn-mediated recycling and increased stability. This platform is a key component of several clinical-stage immunotherapies, most notably nogapendekin alfa inbakicept (N-803), which are being investigated for their ability to stimulate robust anti-tumor immune responses in various malignancies, including bladder cancer and leukemia.
