Interleukin-17F (IL-17F) is a pro-inflammatory cytokine and a member of the IL-17 family, primarily produced by Th17 cells. It functions as a homodimer or a heterodimer with IL-17A, signaling through a heteromeric receptor complex composed of IL-17RA and IL-17RC. While IL-17F is most commonly recognized as a therapeutic target for monoclonal antibodies (such as bimekizumab) in the treatment of inflammatory conditions like psoriasis and hidradenitis suppurativa, recombinant IL-17F is also being investigated as a therapeutic agent. Preclinical research has explored its use in biologic augmentation for rotator cuff tendon healing, where local administration improved fibroblast organization and biomechanical stiffness. Additionally, IL-17F has demonstrated anti-tumor potential in colon cancer models by inhibiting angiogenesis and inducing endothelial cell pyroptosis via the Caspase 4/GSDMD signaling pathway.
