Overview

**interleukin-18 + glycoprotein 100 dendritic cell vaccine** is an experimental cancer immunotherapy that combines *interleukin-18* (IL-18), an immunostimulatory cytokine, with the melanoma-associated antigen *glycoprotein 100* (gp100), delivered via autologous dendritic cells. The vaccine is produced by modifying tumor cells to express IL-18 and incorporating gp100 antigen, then generating dendritic cells pulsed with exosomes from these modified tumor cells. This strategy is designed to enhance dendritic cell maturation, increase the expression of co-stimulatory molecules, and potentiate anti-tumor immune responses—specifically the induction of gp100-specific cytotoxic T lymphocytes (CTLs). Mechanistically, IL-18 acts as a Th1-promoting cytokine that stimulates T cell activation, while dendritic cells present gp100 antigen to provoke specific anti-tumor immunity. The approach aims to improve the efficacy of dendritic cell vaccines for melanoma and other cancers over conventional exosome or antigen vaccines[1].
