**interleukin-18-encoding plasmid DNA** is a form of gene therapy that utilizes a plasmid DNA vector encoding the cytokine interleukin-18 (IL-18). After intratumoral or local delivery, the plasmid leads to local production of IL-18 protein, which is a potent inducer of interferon-gamma (IFN-γ) and enhances cytotoxic and immune-modulating activity of CD4^+ T cells, CD8^+ T cells, and NK cells. This therapy is studied primarily for cancer immunotherapy, especially for solid tumors such as hepatic or colon cancer, aiming to stimulate anti-tumor immunity directly in the tumor microenvironment. The mechanism hinges on the induction of a Th1-type immune response and enhancement of IFN-γ-mediated anti-tumor activity.
