Drug intelligence / Profile preview
Interleukin-2 + anti-interleukin-2 antibody (IL-2/anti-IL-2 complex, or IL-2C) is an immune complex consisting of recombinant interleukin-2 (IL-2) non-covalently bound to a specific anti-IL-2 monoclonal antibody. This approach is widely used in preclinical research to enhance the in vivo half-life of IL-2 and selectively direct its activity toward specific immune cell subsets. Depending on the antibody clone used, the complex can be biased toward different receptors: complexes using clones like JES6-1 (in mice) or F5111.2 (in humans) block the interaction of IL-2 with IL-2Rβ (CD122) and IL-2Rγ (CD132) while favoring binding to IL-2Rα (CD25), thereby selectively expanding immunosuppressive regulatory T cells (Tregs) to treat autoimmune diseases or transplant rejection. Conversely, complexes using clones like S4B6 (in mice) or NARA1 (in humans) block the CD25-binding site, directing IL-2 to the intermediate-affinity IL-2Rβγ complex on CD8+ effector T cells and natural killer (NK) cells to promote antitumor immune responses.
