This is a combination therapy consisting of interleukin-2 (IL-2), a cytokine that stimulates the proliferation and activation of lymphocytes (notably T cells and natural killer cells), and melatonin (MLT), a neurohormone produced by the pineal gland with immunomodulatory properties. The rationale for this combination is based on evidence that melatonin can enhance the biological anti-tumor activity of IL-2, potentially by modulating immune responses through psychoendocrine–immune system interactions. Clinical studies have shown that adding melatonin to low-dose subcutaneous IL-2 increases tumor regression rates and improves survival in patients with advanced solid tumors compared to IL-2 alone. The combination has also been explored in metastatic melanoma as part of chemoneuroimmunotherapy regimens, showing improved efficacy and tolerability[1][3][5].
