Interleukin-2-activated natural killer cells (IL-2 NK) are an adoptive cellular immunotherapy consisting of donor-derived natural killer (NK) cells that are purified and activated ex vivo with high doses of interleukin-2 (IL-2) prior to infusion. This activation process enhances the expression of activating receptors such as NKp44, CD69, and TRAIL, and significantly increases the cells' cytotoxic capacity and cytokine production (e.g., IFN-gamma). In the context of allogeneic hematopoietic stem cell transplantation (allo-HSCT), these cells are administered prophylactically to bolster the graft-versus-tumor (GVT) effect and clear residual disease while minimizing the risk of graft-versus-host disease (GVHD), which is a common complication of conventional donor lymphocyte infusions.
