This drug is a tri-specific immunocytokine consisting of an engineered interleukin-2 variant (IL-2v) fused to a bispecific antibody that targets programmed cell death protein 1 (PD-1) and an additional undisclosed antigen. Developed by ABL Bio using their Grabody-I platform, the IL-2v component is designed with significantly reduced binding affinity for the IL-2 receptor β/γ (IL-2Rβ/γ) and/or α (IL-2Rα) subunits to prevent systemic immune activation and toxicities such as vascular leak syndrome. The anti-PD-1 arm facilitates 'cis-delivery,' ensuring that IL-2 signaling is preferentially restored only upon binding to PD-1-expressing T cells within the tumor microenvironment. This targeted approach aims to overcome resistance to standard PD-1/PD-L1 therapies by providing localized T-cell stimulation while maintaining a favorable therapeutic index.
