Overview

Interleukin-21-enhanced cord blood-derived natural killer (NK) cells are an experimental adoptive cell therapy being developed for the treatment of glioblastoma. These cells are derived from umbilical cord blood and genetically modified (transduced) to express Interleukin-21 (IL-21). This modification triggers metabolic reprogramming and enhances mitochondrial fitness, characterized by improved mitochondrial respiration and CEBPD-driven mitophagy. The resulting NK cells exhibit increased diversity, polyfunctionality, and superior anti-tumor potency against glioblastoma stem cells (GSCs), which are typically resistant to conventional therapies. Preclinical studies have demonstrated that these enhanced NK cells can effectively control tumor growth and improve survival in glioblastoma models without significant toxicity.
