Overview

interleukin-22-expressing Saccharomyces boulardii is a genetically engineered live biotherapeutic product (LBP) developed by researchers at Huashan Hospital and Fudan University. It utilizes the probiotic yeast *Saccharomyces boulardii* as a delivery vehicle to constitutively express and secrete human interleukin-22 (IL-22) directly into the intestinal lumen. IL-22 is a cytokine that plays a pivotal role in mucosal immunity and tissue repair by binding to the IL-22 receptor on intestinal epithelial cells, thereby stimulating the production of antimicrobial peptides (such as Reg3γ) and promoting epithelial cell proliferation and goblet cell differentiation. This localized delivery approach is designed to treat *Clostridioides difficile* infection (CDI) and inflammatory bowel disease (IBD) by restoring the intestinal barrier and modulating the local immune response while minimizing potential systemic side effects associated with systemic cytokine administration.
