Interleukin-24 delE5 (IL-24 delE5) is a naturally occurring splice variant of the mda-7/IL-24 cytokine that lacks the sequence encoded by exon 5. While the full-length mda-7/IL-24 is primarily recognized for its ability to induce apoptosis in a wide range of cancer cells, IL-24 delE5 has been identified as a key regulator of monocytic differentiation in acute myeloid leukemia (AML). It functions by increasing the activity of NADPH oxidase, leading to the generation of reactive oxygen species (ROS), which subsequently triggers the differentiation of leukemic blasts into mature monocytes/macrophages. Research indicates that IL-24 delE5 is induced by 12-O-tetradecanoylphorbol-13-acetate (TPA) through the MAPK/ERK signaling pathway and exhibits selective activity against malignant cells while sparing normal hematopoietic progenitors.
