Overview

Interleukin-25 (IL-25), also known as IL-17E, is a cytokine produced by various cells including eosinophils, T-helper type 2 (Th2) cells, and epithelial cells. It plays a key role in regulating adaptive immune responses by promoting Th2-type immunity. Mechanistically, IL-25 acts via its receptor complex composed of IL17RA and IL17RB to stimulate the JAK2–STAT5A pathway and induce secretion of type 2 cytokines such as IL4, IL5, and IL13. These actions lead to expansion of eosinophils and are implicated in allergic inflammation (e.g., asthma, atopic dermatitis), gut immunity/inflammation (e.g., inflammatory bowel disease), infectious diseases (by modulating IgM/IgE production and pathogen clearance), and antitumor activity in several cancers. Both agonists (recombinant protein) and antagonists/neutralizing antibodies targeting the pathway are under investigation for therapeutic use[2][3][4][5].
