Overview

Recombinant interleukin-30 (rIL30) is a cytokine therapy based on the p28 subunit of interleukin-27. Although p28 typically functions as part of the IL-27 heterodimer, IL-30 can act independently to regulate immune responses. It serves as a natural antagonist of gp130-mediated signaling, which is a shared receptor subunit for the IL-6 family of cytokines. By blocking gp130, IL-30 inhibits the pro-inflammatory signaling of IL-6 and IL-27, affecting the polarization of T helper 1 (Th1) and T helper 17 (Th17) cells. Preclinical research indicates that rIL30 can protect against sepsis-induced mortality by promoting IL-10 production in natural killer T (NKT) cells and reducing systemic inflammation. It also shows potential in treating autoimmune skin conditions like psoriasis by suppressing dendritic cell maturation and keratinocyte cytokine production.
