Overview

pIL30 refers to plasmid DNA encoding interleukin-30 (IL-30), a cytokine subunit also known as IL27p28. The pIL30 construct is designed for gene therapy, where the DNA is delivered (e.g., by electroporation) to cells to induce expression of IL-30 protein. In preclinical models, pIL30 has been shown to protect against sepsis by modulating inflammatory and anti-inflammatory cytokine profiles, particularly through upregulation of IL-10 and downregulation of pro-inflammatory cytokines like IFN-γ and TNF-α[2]. The protective effect is mediated primarily via natural killer T (NKT) cells, which produce IL-10 in response to IL-30 signaling through the gp130 and IL-6 receptor (IL-6R) pathways[2]. IL-30’s effects are independent of its canonical partner EBI3 (which together form IL-27), suggesting a unique pathophysiological role for IL-30[2].
