Interleukin-4 (IL-4) is a pleiotropic cytokine and key regulator in humoral and adaptive immunity. It induces differentiation of naive helper T cells (Th0) to Th2 cells and stimulates activated B cell and T cell proliferation. IL-4 promotes class switching to IgE in B cells and upregulates MHC class II production. It is produced primarily by mast cells, Th2 cells, eosinophils and basophils. Overproduction of IL-4 is associated with allergic diseases such as asthma and atopic dermatitis. Therapeutically targeting or blocking the activity of interleukin-4 has been explored for conditions like atopic dermatitis and asthma; both antibody-based inhibitors (e.g., pasolizumab) and small-molecule inhibitors have been investigated[3][5][8]. Recombinant human interleukin-4 (Binetrakin) has also been studied as an immunomodulatory agent.
