Overview

The interleukin-4 acute myeloid leukemia vaccine is an experimental cancer immunotherapy consisting of acute myeloid leukemia (AML) cells genetically modified to secrete the cytokine interleukin-4 (IL-4). Developed primarily in preclinical research settings, such as at the Dana-Farber Cancer Institute, the vaccine is created by transducing leukemic cells with a retroviral vector (e.g., MFG-IL-4) containing the IL-4 gene. The mechanism of action involves the local production of IL-4 by the tumor cells, which is intended to modulate the immune microenvironment and stimulate a potent anti-leukemic immune response. While preclinical murine models demonstrated that IL-4 secretion could lead to the rejection of the vaccine cells themselves (reduced tumorigenicity), the vaccine was found to be less effective than GM-CSF-transduced vaccines in eliciting systemic, long-term protective or therapeutic immunity against wild-type AML challenge.
