Overview

Interleukin-4 PE38KDEL cytotoxin is a recombinant fusion protein designed as a targeted therapy for cancers that overexpress interleukin-4 receptors (IL-4R). It is composed of a circularly permuted human interleukin-4 (IL-4) component fused to a truncated form of Pseudomonas exotoxin (PE38KDEL). The IL-4 portion specifically binds to IL-4R on cancer cells, facilitating the internalization of the fusion protein. Once inside the cell, the PE38KDEL component, which consists of specific amino acid sequences from Pseudomonas exotoxin, inhibits protein synthesis by ADP ribosylation of elongation factor 2, leading to apoptosis of the cancer cell. This mechanism allows for selective targeting of tumor cells while sparing normal cells that express low or no IL-4R. The drug was developed by Neurocrine Biosciences and produced by Inland Labs.
