Overview

Intestinal epithelial organoid-derived exosomes are nanoscale extracellular vesicles (typically 30–130 nm in diameter) secreted by cultured organoids that recapitulate the structure and function of the intestinal epithelium. These exosomes are characterized by a double-layer membrane and express canonical markers such as CD81 and TSG101[1][3]. They selectively package bioactive proteins, lipids, and nucleic acids reflective of their origin from specific regions of the intestine (e.g., ileum or jejunum), influencing gene expression, metabolic processes, biosynthetic pathways, regulatory functions, transport mechanisms, immune modulation, and intercellular signaling in recipient cells[1][3]. Their biological activities include regulation of inflammation (e.g., suppression of TNF-α and IL-17A), modulation of metabolic pathways (lipid metabolism in ileal IEC-Exos; sugar/fat/protein metabolism in jejunal IEC-Exos), support for tissue regeneration after injury or inflammation, and potential roles in disease models such as colitis or sepsis[1][3].
