A combination investigational regimen composed of three agents: - **Intismeran autogene** (also known as V940, formerly mRNA-4157) is an individualized, patient-specific neoantigen therapy based on mRNA technology. It is designed to stimulate immune responses targeting patient-specific tumor neoantigens. - **Pembrolizumab** is a humanized monoclonal antibody that inhibits the programmed cell death protein 1 (PD-1) pathway, which enhances the immune system's ability to detect and attack tumor cells. - **Enfortumab vedotin** is an antibody-drug conjugate consisting of a monoclonal antibody directed against Nectin-4, a protein highly expressed on some tumor cells, conjugated to the microtubule-disrupting agent monomethyl auristatin E (MMAE). This combination is intended for use in certain high-risk or advanced cancers, including melanoma and potentially other solid tumors, to combine individualized tumor antigen vaccination, immune checkpoint inhibition, and direct tumor cell killing by ADCs[1][2][3][4][5][6].
