Overview

Intranasal insulin is a delivery method that administers insulin through the nasal cavity, allowing it to reach the central nervous system while minimizing systemic absorption and peripheral side effects. It rapidly accumulates in cerebrospinal fluid, indicating efficient transport to the brain. This administration route enables insulin to exert functional effects on metabolism (reducing food intake and body weight, improving glucose homeostasis) and cognition (enhancing memory performance in both healthy individuals and patients with mild cognitive impairment or Alzheimer's disease). Intranasal insulin also modulates neuroendocrine activity, sleep-related mechanisms, sensory perception, and mood. It is being investigated as a potential treatment for cognitive disorders, particularly Alzheimer's disease, based on the concept that impairments in central nervous insulin signaling may be a pathophysiological factor in metabolic and cognitive disorders.
