Overview

Methylprednisolone is a synthetic glucocorticoid corticosteroid being investigated by Ziv Hospital for the prevention of cisplatin-induced ototoxicity in adult cancer patients. Cisplatin, a common chemotherapeutic agent, often causes irreversible, bilateral, high-frequency sensorineural hearing loss by inducing reactive oxygen species (ROS) formation and cellular apoptosis in the inner ear. By administering methylprednisolone locally via intratympanic injection, researchers aim to achieve high concentrations of the steroid in the perilymph of the inner ear to activate glucocorticoid receptors, thereby exerting anti-inflammatory and anti-apoptotic effects while minimizing systemic side effects. This specific intervention (NCT01285674) involves a 62.5 mg/cc concentration injected before cisplatin cycles.
