InvaplexAR-Detox is a second-generation, subunit vaccine candidate designed to protect against *Shigella* infection, the primary cause of bacillary dysentery. It is a 'detoxified' version of the Artificial Shigella Invaplex (InvaplexAR), which is a synthetic macromolecular complex composed of highly purified *Shigella* invasins (IpaB and IpaC) and lipopolysaccharide (LPS). The 'Detox' version specifically utilizes a detoxified form of LPS (such as de-acylated LPS or LPS from modified strains) to reduce the reactogenicity and potential side effects associated with native endotoxin while maintaining the potent adjuvant-like properties and immunogenicity of the complex. Developed primarily by researchers at the Walter Reed Army Institute of Research (WRAIR), the vaccine is designed to be administered intranasally or parentally to induce both mucosal and systemic immunity, targeting the essential virulence machinery of the pathogen to prevent bacterial entry into host epithelial cells.
