Drug intelligence / Profile preview
Invikafusp alfa (STAR0602) in combination with sacituzumab govitecan (Trodelvy) is an investigational combination regimen being evaluated for the treatment of advanced solid tumors, specifically unresectable, locally advanced, or metastatic breast cancer (including triple-negative breast cancer and HR+/HER2- breast cancer). Invikafusp alfa is a first-in-class, bispecific dual T-cell agonist designed to selectively target and activate Vβ6 and Vβ10 T-cell receptor (TCR) subsets, promoting the expansion of clonally diverse effector memory T cells. Sacituzumab govitecan is a TROP2-directed antibody-drug conjugate (ADC) carrying a topoisomerase I inhibitor payload (SN-38) that induces immunogenic cell death and releases tumor-associated antigens. The combination leverages the ADC's tumor-killing and antigen-releasing properties to prime the tumor microenvironment, enhancing the anti-tumor activity of the selectively activated T cells. The regimen is being developed by Marengo Therapeutics in collaboration with Gilead Sciences.
