Inzomelid is an oral, brain-penetrant small molecule inhibitor of the NLRP3 inflammasome (NOD-like receptor family, pyrin domain-containing protein 3). It was originally developed by Inflazome and later acquired by Roche. Inzomelid is designed to selectively inhibit activation of the NLRP3 inflammasome, a multiprotein complex involved in innate immune responses and chronic inflammation. Activation of this inflammasome leads to production of pro-inflammatory cytokines IL1β and IL18, contributing to diseases such as Cryopyrin-associated periodic syndromes (CAPS), Alzheimer's disease, and Parkinson's disease. Inzomelid has shown positive results in early clinical trials for CAPS and received orphan drug designation from the FDA for this indication[1][3][5][6].
