Overview

IO-108 + cemiplimab is a combination of two monoclonal antibodies under investigation for the treatment of advanced solid tumors and other cancers. IO-108 is a fully human IgG4 monoclonal antibody that targets and blocks leukocyte immunoglobulin-like receptor B2 (LILRB2, also known as ILT4), a myeloid checkpoint involved in cancer-associated immune suppression. By inhibiting LILRB2, IO-108 aims to promote both innate and adaptive anti-cancer immunity. Cemiplimab is an anti-PD-1 monoclonal antibody (marketed as Libtayo) that blocks the programmed cell death protein 1 (PD‑1) on T cells, thereby enhancing T cell-mediated immune responses against tumor cells. The combination seeks to overcome resistance mechanisms in tumors by targeting both myeloid and T cell checkpoints, with clinical trials showing encouraging safety profiles and preliminary efficacy signals in various solid tumor types[1][2][5][7][8].
