IO-108 + pembrolizumab

IO-108 is a fully human IgG4 monoclonal antibody that targets leukocyte immunoglobulin-like receptor B2 (LILRB2, also known as ILT4), a myeloid checkpoint molecule. Pembrolizumab is a monoclonal antibody targeting programmed cell death protein 1 (PD-1). The combination of IO-108 and pembrolizumab is being investigated for the treatment of advanced solid tumors, including hepatocellular carcinoma and other relapsed or refractory solid tumors. In clinical studies, this combination has demonstrated safety and preliminary efficacy, with an overall response rate of 23% in early-phase trials for patients with advanced solid tumors who had failed prior therapies. The rationale for combining these agents is to reduce immune suppression by blocking both myeloid (LILRB2) and lymphoid (PD-1) checkpoints, thereby enhancing antitumor immune responses[2][3][4][5].
