IO-108 + tislelizumab is a combination of two monoclonal antibodies under investigation for the treatment of advanced solid tumors, including hepatocellular carcinoma and other cancers. IO-108 is a fully human IgG4 monoclonal antibody that targets the myeloid checkpoint receptor Leukocyte immunoglobulin-like receptor subfamily B member 2 (LILRB2) (also known as ILT4), blocking its interaction with ligands such as HLA-G, ANGPTLs, SEMA4A, and CD1d to relieve cancer-associated immune suppression and enhance T-cell activation. Tislelizumab is a humanized IgG4 monoclonal antibody targeting Programmed cell death protein 1 (PD-1), blocking PD-L1/PD-L2-mediated signaling to restore T-cell function and promote antitumor immune responses. The combination aims to potentiate immune activation by simultaneously inhibiting myeloid-derived suppressive pathways (via LILRB2) and adaptive immune checkpoints (via PD-1). Clinical trials have shown that this combination is well tolerated with encouraging clinical activity in patients with advanced solid tumors[1][3][5][6].
