Overview

IO-202 analog is a research-grade monoclonal antibody designed to replicate the binding and functional characteristics of IO-202, a first-in-class therapeutic antibody targeting Leukocyte Immunoglobulin-Like Receptor B4 (LILRB4). LILRB4 is an inhibitory immune checkpoint receptor that is highly expressed on monocytic acute myeloid leukemia (AML) cells and other myelomonocytic lineages. By binding to LILRB4, the analog blocks its interaction with the ligand Apolipoprotein E (APOE), thereby preventing the activation of SHP-1/2 signaling pathways that facilitate leukemia cell infiltration and immune evasion. In preclinical research, IO-202 analogs are used as reference standards to evaluate the efficacy of novel LILRB4-targeted agents, such as BCG042, in assays measuring signaling inhibition, antibody-dependent cellular cytotoxicity (ADCC), and in vivo tumor regression.
