Overview

IO-312 is a first-in-class bispecific monoclonal antibody engineered to target LILRB4 (also known as ILT3) and CD3. Developed by Immune-Onc Therapeutics, it is designed for the treatment of acute myeloid leukemia (AML) and other hematologic malignancies. The drug binds with high affinity to LILRB4 and with low affinity to CD3, aiming to maximize anti-tumor effects while minimizing cytokine release-related toxicities. Preclinical data demonstrate potent and specific killing of monocytic AML cells both in vitro and in vivo. IO-312’s design allows for efficient manufacturing and cross-reactivity with non-human primate targets, supporting preclinical toxicology studies[1][3].
