Overview

IO102 is a second-generation peptide-based cancer vaccine developed as an immunotherapy targeting the enzyme indoleamine 2,3-dioxygenase (IDO). It is designed to stimulate cytotoxic T lymphocyte responses against IDO-expressing cells in the tumor microenvironment. By inhibiting IDO—a key immunosuppressive enzyme often overexpressed in various cancers—IO102 aims to reduce immune suppression and enhance anti-tumor immunity. The drug is being investigated primarily in combination with other therapies such as pembrolizumab (an anti-PD-1 antibody) and sometimes with another peptide vaccine (IO103), which targets PD-L1 positive cells. Clinical trials have focused on advanced melanoma, squamous cell carcinoma of the head and neck (SCCHN), and non-small cell lung cancer (NSCLC). IO102 is administered subcutaneously.
