IO102 + IO103 + pembrolizumab is a combination immunotherapy regimen under clinical investigation for the treatment of advanced cancers. It consists of two investigational therapeutic cancer vaccines (IO102 and IO103) and the anti-PD-1 monoclonal antibody pembrolizumab (marketed as Keytruda). - **IO102** targets indoleamine 2,3-dioxygenase (IDO), an enzyme that suppresses immune responses in the tumor microenvironment. - **IO103** targets programmed death-ligand 1 (PD-L1), another key immune checkpoint molecule expressed by tumor cells and immune-suppressive cells. Both vaccines are designed to stimulate activation and expansion of T cells against IDO-positive and/or PD-L1-positive cells, thereby enhancing anti-tumor immunity. Pembrolizumab blocks PD-1 on T-cells to prevent inhibition by PD-L1/PD-L2 on tumor or other cells, further promoting T-cell mediated anti-tumor activity[4][9]. The combination is being evaluated as first-line therapy for advanced melanoma, metastatic non-small cell lung cancer (NSCLC), recurrent/metastatic squamous cell carcinoma of the head and neck (SCCHN), among others[4][5][6].
