Overview

IO102 + Montanide is an investigational therapeutic cancer vaccine combination consisting of a 21-amino acid peptide derived from the Indoleamine 2,3-dioxygenase (IDO) enzyme, emulsified with the water-in-oil adjuvant Montanide ISA 51. Developed by IO Biotech, IO102 is designed to stimulate the immune system to recognize and eliminate IDO-expressing cells, which include both malignant tumor cells and immunosuppressive cells (such as regulatory T cells and myeloid-derived suppressor cells) within the tumor microenvironment. IDO is a key metabolic checkpoint that suppresses T-cell activity by depleting tryptophan and producing kynurenine. By inducing IDO-specific cytotoxic T-lymphocytes (CTLs), the vaccine aims to reverse local immunosuppression and enhance anti-tumor immunity. It is primarily being evaluated in squamous cell carcinoma of the head and neck (SCCHN) and other solid tumors, often in combination with immune checkpoint inhibitors like nivolumab or pembrolizumab.
